Fulham were scheduled to be back in action at Turf Moor on Sunday, with kick-off set for midday.
But the match has been called off after the latest batch of testing revealed more positive results.
A statement from Fulham read: “Following further new positive results returned from the most recent Covid-19 tests, the Club can confirm that this Sunday’s away at fixture at Burnley has been postponed after discussions with the Premier League and advice from the medical teams.
“Those who tested positive are self-isolating in line with UK Government and Premier League guidance.
“The welfare of our players, staff and their families continue to be of paramount importance, and we wish those who have tested positive a safe and speedy recovery.”
Fulham boss Parker missed his club’s Boxing Day meeting with Southampton.
The Cottagers boss has tested negative for Covid-19, but had to isolate after a member of his household returned a positive test.
This is the fourth game of the 2020-21 season to be postponed following a surge in coronavirus cases before kick-off.
Aston Villa’s game against Newcastle and Manchester City’s meeting with Everton have also been postponed.
Despite the latest Covid spike, Premier League bosses are determined to keep the season on track.
There are increasing concerns over the spread of the virus after the discovery of the fast-growing mutant strains.
But the top-flight officials are still said to have ‘full confidence’ in the protocols that are in place.
Fulham and Crystal Palace are investigating after a video emerged of their stars appearing to break Covid rules.
Aleksandar Mitrovic and Luka Milivojevic were filmed together on Instagram by the Fulham striker’s long-term partner Kristina Janjic at a London New Year party.
Then a later story posted shows Mitrovic playing football with Eagles captain Milivojevic as well as another man – who appears in the earlier video – and two children wearing this season’s Fulham kits.
Meanwhile, Spurs have condemned three stars after they were accused of breaking coronavirus guidelines to hold a Christmas party together at Giovani Lo Celso’s home.
