Ajax took the lead in the first half through Davy Klaassen, and had the chance to double their advantage after the break but Dusan Tadic missed from the spot after he was brought down in the box.
Roma equalised through Lorenzo Pellegrini and snatched it late on through a stunning volley from Roger Ibanez.
The second leg will take place in Rome on Thursday, 15th April at 8pm.
